O’Sullivan, 48, knocked in breaks of 129, 106 on the way to going 3-1 up in Yushan before Michael White levelled at 3-3.
An 87 put world number one O’Sullivan back in front and he took the eighth frame to claim victory.
World number two Trump’s highest break was 73 as he secured a 5-2 win against Sanderson Lam.
England’s Trump won the tournament last time it was held in 2019 and will hope to defend that title following its return to the tour after a four-year hiatus because of the coronavirus pandemic.
World champion Luca Brecel beat Oliver Brown 5-2, while home favourite Ding Junhui won by the same score against compatriot Liu Hongyu.
Four-time world champion John Higgins is out of the tournament following a 5-3 defeat by Jackson Page.
